Zach Putnam struck out two in the eighth and Javy Guerra fanned two more in the ninth.
Alexei Ramirez hit a two-run homer for the White Sox, while Andy Wilkins drove in the first run of his career and Jose Abreu doubled twice in the win.
Six of Chicago's seven hits went for extra bases.
Twins starter Phil Hughes (15-10) took the loss after giving up all five runs -- three earned -- on six hits in seven innings. He struck out 11.
Carlos Sanchez reached safely on Joe Mauer's third-inning error around strikeouts by Wilkins and Adam Eaton. Ramirez then crushed the first pitch he saw into the seats in left for his 15th homer of the year.
The White Sox padded their lead in the home seventh, when Jordan Danks doubled and Wilkins drove him in two batters later with a single to center.
Quintana went back out for the eighth inning but walked Kurt Suzuki. Putnam took over and pinch-hitter Chris Parmelee roped a double to deep center off him, plating Suzuki for Minnesota's only run.
