Ezequiel Carrera and Kosuke Fukudome added two RBI apiece and Shelley Duncan chipped in with two hits, including a double for the Indians, who snapped a four-game skid.
Jeanmar Gomez (3-2) tossed six strong innings, yielding two runs on six hits while fanning three without issuing a walk.
Gordon Beckham went 2-for-4 with a double and Juan Pierre smacked an RBI double for the White Sox, who failed to carry over momentum from an 8-1 rout on Thursday.
Mark Buherle (11-8) was tagged with the loss after giving up seven runs on eight hits in 5 2/3 innings. He struck out four and walked two.
The Indians opened the scoring in the second inning when Duncan singled to center to lead off and Chisenhall followed by blasting an offering from Buehrle over the wall in right for his fifth homer of the season.
"He's a guy that projects not to be a platoon guy or a guy whose going to struggle against lefties," Indians manager Manny Acta said of Chisenhall. "His swing is so short, no holes in it."
Beckham slapped a single to get things started in the third and moved to second on a wild pitch by Gomez before coming home with Chicago's first run on a double into the left-center field gap by Pierre. He scored on a sacrifice fly by Paul Konerko.
The Indians broke the game open in the sixth with five runs. Santana worked a one-out walk and Chisenhall cracked his second homer of the game over the right field wall to give the Indians a two-run lead and Carrera doubled home Lou Marson later in the frame to make it 5-2. A two-run single by Fukudome made it 7-2.
"He hung a couple pitches and was kind of erratic," White Sox manager Ozzie Guillen said of his starter's performance. "I think the big hit was when Carrera got that double, that one killed him. Even when he was getting outs, guys were hitting the ball pretty hard."
Cleveland added another run in the eighth inning when Trevor Crowe walked with one out, stole second and hustled home on a double to right-center field by Carrera.
The White Sox got a run back in the home half of the inning when Beckham doubled to lead off the frame, moved to third on a flyout by Pierre and scored on a groundout by Ramirez.
In the ninth, Dayan Viciedo worked a one out walk against Nick Hagadone. Following an Alejandro De Aza foul out, Hagadone plunked Tyler Flowers and walked Brent Morel to load the bases with two down. Viciendo scored on a passed ball by Marson to account for the final margin, but Beckham was caught looking at strike three to end the game.
