Griffin needed less than 31 minutes to post 27 points, nine rebounds and eight assists while Chris Paul tallied 24 and 11 assists in just over 31 minutes.
"We had a good mindset coming into the game," said Griffin.
All five Clippers starters reached double figures, with Matt Barnes scoring 19 points and DeAndre Jordan and J.J. Redick finishing with 10 apiece.
The Clippers have now won five in a row against the Lakers since a 116-103 loss on opening night last season.
Jordan Hill and Wayne Ellington both had 13 points for the Lakers. Kobe Bryant finished 2-for-12 shooting for a season-low four points. He also had teams highs of eight rebounds, seven assists and six turnovers.
Bryant missed Monday's loss in Portland, as well as three games in late December, to rest his body.
"It's very disappointing," said Lakers coach Byron Scott. "We came out soft. They punched us. I told them to man up. Bottom line, we were soft."
The Clippers won each of the first three quarters by 10-plus points. They led 34-24 after one then shot 14-for-22 in the second to extend the spread to 70-48 at intermission.
Barnes capped a 15-3 run to open the third with a 3-pointer from Paul. The lead, which was at 34 points following Barnes' triple of the contest, reached 37 later in the stanza and 43 early in the fourth.
The Lakers made the final score look more respectable, outscoring the Clips 26-15 in the fourth quarter. They finished the game on a 26-8 run.

The Lakers outrebounded the Clips, 46-42. Jordan had a game-high 13 boards ... Lakers rookie Jordan Clarkson scored 10 of his team-high 14 points in the fourth quarter.
