Marcus Morris had a team-high 16 points to go with 11 caroms for the Jayhawks (24-1, 9-1 Big 12), who have rattled off six straight wins since falling to Texas.
Tyshawn Taylor totaled 14 points and six assists, while Brady Morningstar contributed 13 points in the runaway decision.
Kansas, which shot 52 percent from the field, may slide into the No. 1 spot when the rankings come out on Monday as a result of top-ranked Ohio State falling at Wisconsin earlier Saturday.
"I want to be ranked number one," Marcus Morris said. "I want to have that chip on our shoulder every time we go out. I want to get everybody's best shot because I think we can take it."
Scott Christopherson scored 16 points to lead the Cyclones (14-11, 1-9), losers of seven in a row.
The Jayhawks extended an early 20-12 lead with a balanced 13-0 run. Taylor ignited the six-minute stretch with a jumper, and a Marcus Morris free throw with 6:28 left in the half yielded a 33-12 cushion.
Jamie Vanderbeken, who scored seven of Iowa State's first 12 points, injured his ankle during the run to add injury to insult. He did not return.
"Right now it looks like a high-ankle sprain," Iowa State head coach Fred Hoiberg said of Vanderbeken's injury. "He was pretty swollen and he got hurt pretty good. It's a tough blow obviously."
Morningstar's bucket in the final minute capped a first half that saw Kansas shoot 56 percent from the field en route to a healthy 45-26 advantage.
The Cyclones never seriously threatened in the final 20 minutes.
