Alejandro De Aza added a two-run homer, while Paul Konerko had two hits and a pair of RBI for the White Sox, who swept the weather-shortened two-game series.
John Danks (1-1) allowed four runs on seven hits and five walks in 5 2/3 innings.
"It was a good win, it was good to go 3-2 on the road trip," Pierzynski said.
Travis Hafner had three RBI, including a solo shot for Cleveland, which is off to a 1-4 start.
"Regardless of the result I thought the homestand could've gone a little better," Cleveland manager Manny Acta said. "We put ourselves in position to win a couple games and things just didn't work out."
Justin Masterson (0-1) was touched for five runs -- three earned -- on eight hits and one walk in five frames.
Chicago broke the game open with a five-run sixth.
After Gordon Beckham laced a one-out double, De Aza launched a two-run shot to deep center. Brent Morel walked and following Adam Dunn's groundout, Rafael Perez intentionally put Konerko on. Pierzynski stepped up next and cleared the bases with a three-run homer to right field, giving the White Sox a commanding 10-4 advantage.
Shelley Duncan hammered a two-run homer to left field in the seventh to cut the margin to 10-6.
The White Sox jumped on Masterson and the Indians early with four runs in the first. De Aza led off with a single and Jack Hannahan committed a throwing error to first on Morel's sacrifice bunt.
Dunn slapped a single up the middle to bring home De Aza and Konerko laced a grounder just inside the third-base line for a double, plating Morel. Dunn then scored on Masterson's wild pitch and Pierzynski lifted a sac fly to center field to bring home Konerko.
Asdrubal Cabrera doubled and scored on a wild pitch in the bottom of the first and Hafner crushed a hanging breaking ball into the second deck in right field to pull Cleveland within 4-2 in the fourth.
Morel reached on Casey Kotchman's error at first base and scored two batters later on Konerko's single through the hole on the left side to make it 5-2 White Sox in the fifth.
Cleveland, though, got two runs back in the bottom half of the frame. After a pair of one-out walks to Shin-Soo Choo and Carlos Santana, Hafner doubled to right field to bring them home, making it 5-4.
Game Notes
Cleveland was 1-for-11 with runners in scoring position while Chicago was 6- for-9...Konerko is 9-for-20 to start the season with five runs batted in.
