Mike Conley was unable to hit on a pair of potentially game-tying 3-point attempts in the final 10.7 seconds, the latter which rimmed out at the horn.
Walker was 9-of-20 from the field and made all 10 of his free throws for Charlotte, which has won four straight.
"To win four in a row against good teams and to play this well in a game like that that was hard fought, you have to feel good about it," Bobcats head coach Steve Clifford said.
Josh McRoberts was the only other Bobcat in double figures with 12.
Conley finished with 16 points, five assists and five boards, while Zach Randolph added 12 points and 11 rebounds for the Grizzlies, who had their four-game winning streak snapped.
"We wanted to win, but the Charlotte Bobcats were the aggressors," Grizzlies guard Tony Allen said.
After Conley split a pair of free throws to get the Grizzlies within 71-69 early in the fourth, Anthony Tolliver nailed a pair of 3-pointers during an 11-0 Charlotte run, including one to cap it for an 82-69 advantage with 7:19 left.
Memphis countered with an 18-4 surge as Conley's left elbow jumper staked the visitors to an 87-86 lead with 50.1 ticks to play.
Walker drove into the paint and hit a cutting McRoberts along the left baseline and he drew a foul. He hit both free throws to put Charlotte back up by a point with 38.1 seconds left.
Gerald Henderson outbattled Randolph underneath for a key rebound following a Conley airball from the left wing. Walker then hit a pair of foul shots to put the Bobcats up three.
Gasol's layup at the other end whittled to gap to 90-89, but Walker again hit two foul shots for a 92-89 advantage with 10.7 seconds remaining.
Earlier, a 10-0 spurt late in the opening frame propelled Charlotte to a 28-21 margin after a quarter of play, but Memphis outscored the Bobcats 27-14 in the second and held a 48-42 edge at the break.
McRoberts netted seven points during a 15-4 burst to begin the third period. Henderson finished off the flurry with a jumper and a 57-52 Charlotte advantage midway through the stanza.
Game Notes
Charlotte shot just 36.6 percent (30-of-82) from the field, but 88.9 percent (24-of-27) from the foul line ... The Bobcats netted 20 points off 15 Memphis turnovers.
