LeBron James, playing in his second game since breaking his nose, scored 20 points to go along with nine rebounds and seven assists. The Heat have won nine straight over the Magic and swept the season series for the second consecutive year.
Chris Bosh posted 17 points in the Heat's seventh consecutive victory overall.
"I'm in one of those grooves right now," said Wade. "(I'm) taking my shots, some of them are wide-open, some are not, but I'm just taking the shots that I'm comfortable with."
Tobias Harris finished with 20 points, while Nikola Vucevic recorded an 18- point, 10-rebound double-double in the setback.
Kyle O'Quinn contributed 14 points with a career-high 15 rebounds. Orlando couldn't build off of Wednesday's road win over Philadelphia, which snapped a 16-game skid away from home for the Magic.
"Anytime when you're playing the best teams in the NBA, defending champions, you really have to be on your game," said Harris.
With the game tied at 33-33 early in the second quarter, Wade scored six straight to give the Heat an edge with just under 10 minutes remaining.
Norris Cole and Greg Oden each made a pair of buckets during a 9-2 spurt later in the frame to give the Heat a 56-45 lead, but Orlando cut into the deficit and trailed 61-53 at the break.
Miami maintained a healthy advantage for much of the third quarter, as the Heat stretched their lead to 20, 89-69, following an 11-2 spurt late in the stanza.
Orlando couldn't get any closer than 12 in the final 12 minutes as the Heat cruised to a victory.
Earlier, the first quarter was a tightly contested affair with the Heat going up by as many as seven and Orlando taking a brief lead of three. There was one tie with eight lead changes and the Heat brought a 32-30 advantage into the second frame.
