Jamal Crawford scored 29 points in 29 minutes off the bench and DeAndre Jordan had 11 points, 16 rebounds and three blocks for the Clippers, who were coming off a 4-0 homestand.
They won their fourth in a row over the Knicks and opened a seven-game road trip that continues against Indiana on Saturday.
They are 5-1 since losing point guard Chris Paul to a separated shoulder.
"We buckled down and played within ourselves and really relied on our system," said Griffin. "No one is trying to do too much."
Carmelo Anthony had 26 points and 20 rebounds and J.R. Smith scored 24 off the bench to lead the Knicks, who lost their third straight to open an eight-game homestand. It was Anthony's 15th double-double of the season.
The Clippers outscored the Knicks 34-7 on the fast break and scored 23 points off 20 New York turnovers.
It was 50-50 at halftime but the Knicks made just four of their 22 shots in the third quarter while the Clippers shot 50 percent to take a 76-67 lead into the fourth.
Five players scored during a 12-0 Clippers run in the quarter highlighted by Griffin's three-point play on an alley-oop dunk.
LA's lead was in double digits for the last 10:33 of the game and reached 19 points twice.
"Once they started executing their stuff and went on a little run, it was hard for us to bounce back and we couldn't turn the corner," said Anthony.
The Clippers shot 42.3 percent for the game and the Knicks just 34.9 percent, though they were hotter early in the game.
The Knicks shot better than 52 percent in the first quarter and led by as many as eight at the end of a 9-0 run keyed by an Anthony 3. The Clippers scored 14 of their 23 points on fast breaks and were within four at the buzzer.
An 8-0 run gave the Clippers the lead in the second quarter. They went up by four on a two-handed alley-oop dunk by Griffin, who took off midway between the foul line and the rim to catch Crawford's hook-shot pass from the low right side.
Game Notes
Hedo Turkoglu, signed by the Clippers on Thursday, made his only shot in 6:43 off the bench. Turkoglu was cut by Orlando two weeks ago without appearing in a game this season. He hadn't played in a game since last Feb. 4 ... The Knicks will host city rival Brooklyn on Monday.
